AI Helps Develop Cancer Vaccine That Passed Phase 3 Trials

Moderna and Merck announced that their personalized mRNA melanoma therapy successfully completed phase 3 trials, marking a significant milestone in the development of neoantigen vaccines. The use of AI algorithms for predicting immunogenicity played a crucial role in this achievement.
AI Helps Develop Cancer Vaccine That Passed Phase 3 Trials
On August 19, Moderna and Merck announced that their personalized mRNA melanoma therapy successfully completed the first phase 3 trials in history for neoantigen vaccines. Prior to this, such drugs had never been tested on such a sample in a randomized study. Thus, this is the first rigorous evidence that the approach actually works.
The treatment is based on AI algorithms for predicting immunogenicity. The AI algorithm from Moderna compares tumor DNA with healthy tissue and predicts which of the thousands of mutations will yield peptides capable of binding to HLA and "triggering" T-cells. The accuracy was confirmed on archival data: the model correctly identified the neoantigens that T-cells from patients actually responded to. From the selected targets, a single mRNA is created for 34 neoantigens and injected into the patient.
The phase 3 figures have not yet been disclosed, but in phase 2, a 49% reduction in the risk of recurrence or death and a 59% reduction in the risk of metastasis were observed compared to Keytruda (one of the best-selling drugs in the world).
Next, the vaccine will begin testing on tumors in the lungs, kidneys, and others.
Why it matters
AnalysisThis is significant as the successful completion of phase 3 trials for neoantigen vaccines could change cancer treatment approaches, opening new possibilities for personalized medicine. The use of AI in this field demonstrates the potential of technology to improve treatment outcomes.
